Protein Purification via Crossflow Filtration

Crossflow filtration, also known tangential flow filtration, is an important technique in protein purification and the manufacturing of pharmaceutical products. Unlike conventional, so called dead-end filtration, where the solids are trapped in a filter and the filtrate passes through, with crossflow filtration the feed solution is passed over the filter at positive pressure. The size of permeate produced is therefore a product of the filter pore size employed and the process can become continuous (through recycling) as the filter does not become readily ‘caked'. Crossflow filtration is especially applicable in protein purification and where high-value small molecules need to be purified, hence its importance in the production of blood factors, IgG, peptides, enzymes and related drug products.
Crossflow ‘cassettes' are an effective and effecient means of carrying out crossflow filtration and Sartorius with their compact SARTOFLOW® Advanced have produced a modular benchtop cassette system that can concentrate even very small process batches. And because the SARTOFLOW® Advanced will accept either Sartocon® Slice or full-sized Sartocon® cassettes a wide range of membrane surface areas becomes available. In practice, because all Sartocon® cassettes have the same hydrodynamic flow path length, it means that the SARTOFLOW® Advanced can be linearly scaled up from 0.1 to 2.1 m2 of membrane area.
This makes the SARTOFLOW® Advanced eminently suitable for the different microfiltration, ultrafiltration and diafiltration procedures involved in the purification of vaccines, monoclonal antibodies and recombinant proteins and hence for small-scale batch production as well as clinical trials and process development.
The whole unit has in fact been designed especially to withstand the rigours of continuous usage such jobs might well demand. And because the SARTOFLOW® Advanced has a proven DCU-4 control unit, which readily communicates with Sartorius´ BioPAT® SCADA, MFCS software, protein purification has really never been easier, whatever your application.
